Comprehensive Review of Fundamental Biochemical Techniques: Integration of Quantification, Separation, and Analytical Methods in Biomolecular Studies
Year: 2024
Type: article
Abstract: This project includes nine experiments that explore important concepts and techniques in biochemistry. We started with basic laboratory practices, such as preparing different solutions (molar, molal, and normal) and learning about dilution, density, specific gravity, and limiting reagents. We then determined the absorption maxima (λmax) of a solution to study its light absorption and performed a titration of a weak acid with a strong base to understand acid-base reactions. Quantitative estimation of carbohydrates using dinitrosalicylic acid helped us measure sugar content, while Lowry’s method was used to estimate the amount of protein in a sample. Other experiments included identifying reducing and non-reducing sugars, analyzing DNA to check its concentration and purity, and separating amino acids using paper and thin-layer chromatography. These experiments taught us how to work with biomolecules and use various biochemical techniques effectively.
