ValveCCI-seq: A High-throughput, Broadly Compatible Microfluidic Platform for in Situ Cell-Cell Interaction Sequencing
Year: 2025
Type: article
Abstract: Immunotherapy achieves the selective eradication of diseased cells by precisely activating or enhancing the specific responses of the patient's immune system. The efficacy of immunotherapeutic approaches fundamentally relies on the precise modulation of immune cell interaction networks with disease-relevant cellular targets. Current methodologies for analyzing cell-cell interactions face irreconcilable trade-offs between pairing throughput, efficiency, and cellular compatibility, which pose substantial limitations, particularly when working with high-value clinical specimens. To address these limitations, we have developed a microfluidic platform that integrates deterministic cell pairing module with in-situ sequencing module for the investigation of cell interaction. ValveCCI-seq analysis of Jurkat and T2 cells under different conditions revealed gene expression profiles associated with T-cell activation, thereby demonstrating the platform's capability for the precise identification of tumor antigens and their corresponding specific T-cell receptor (TCR) sequences. In summary, ValveCCI-seq serves as a potent methodology for elucidating the mechanisms underlying cellular interactions, enhancing the efficacy of cell-based therapies, and identifying novel targets for pharmacological intervention.
