Abstract: Alzheimer's disease is a syndrome characterized by cognitive impairment, behavioral disturbances and loss of autonomy in daily life. It mainly occurs after the age of 65. The prevalence of Alzheimer's disease is increasing dramatically due to the aging of the population. It is therefore a growing public health crisis. Cholinesterase inhibitors and memantine are symptomatic treatments of Alzheimer's disease. They have no effect on the progression of cognitive decline. The intensification of clinical research is crucial due to the lack of therapies capable of slowing down or preventing the deterioration of intellectual functions. Dozens of drugs are currently in clinical trials and some of them are likely to be marketed in the years to come. Two disease-modifying treatments have been approved in the United States in 2021 and 2023 for the treatment of Alzheimer's disease. These are anti-amyloid antibodies : aducanumab and lecanemab. A third anti-amyloid antibody, donanemab, is being assessed in phase III clinical trials to be commercialized in the short term. In addition to anti-amyloid and anti-Tau medication, drugs targeting neuroinflammation or disruption of gut microbiota are now candidates for marketing authorization. An effective disease-modifying treatment of Alzheimer's disease will likely be a drug combination therapy including anti-amyloids, anti-Tau, immunomodulators and neuroprotective compounds. Hence, drug approval in Europe seems possible and would be an unprecedented source of hope for millions of people.
