Insulin signaling regulates localization and translation of Pink1 mRNA via modulation of AMPK activity to support mitophagy in neurons

Year: 2023
Type: dissertation
Abstract: Neurons’ extended structure challenges mitochondrial maintenance. PINK1-mediated mitophagy prevents accumulation of defective mitochondria, vital to prevent neurodegeneration. Pink1 mRNA travels with mitochondria via SYNJ2BP/SYNJ2 allowing for on-demand local translation. AMPK phosphorylates SYNJ2BP at S21. Insulin-mediated inhibition of AMPK reduces mitochondrial Pink1 mRNA tethering, which increases its translation and promotes mitophagy. ApoE4-induced insulin resistance disrupts PINK1, linking neurodegeneration, mitochondrial dysfunction and insulin.
