Customized DNA origami for delivery of therapeutic cargoes.

Year: 2022
Type: conference-paper
Abstract: DNA is an attractive molecular building block to construct nanoscale structures for several applications. In addition to their unique structure, the conjugation and chemical modification of DNA nanostructures with other molecules opens unlimited possibilities in the production of functional DNA-based architectures. For therapeutic applications, one of the challenges regards to the internalization of DNA scaffolds into cells for an efficient drug delivery. \nHere, we exploited the possibility of using two lipids, cholesterol and oleic acid, as potential enhancers of DNA origami¿s cellular uptake. We demonstrate that the incorporation of oligonucleotide-lipid conjugates into DNA origami not only does not disrupt DNA nanostructure formation, but increases its cellular uptake compare with non-modified DNA origami. These results suggest an improvement in the application of DNA nanostructures as drug delivery vehicles. Moreover, 5-fluoro-2¿-deoxyuridine (FdUn) oligomers were incorporated in DNA origami revealing an ability to circumvent 5-FU sensitivity of colorectal drug-resistant cancer cells and exhibiting promising results regarding cytotoxicity in two different colorectal cancer cells. \nThe present work paves the way for achieving a higher cell internalization of DNA origami and confirms it as a privileged carrier for delivering drugs, opening new promising avenues for the development of novel therapeutic systems for cancer treatment.
