Structural analysis of short-term release facilitation in synaptosomes and synapses
Year: 2018
Type: dissertation
Abstract: By means of cryo-electron tomography, native synaptosomes and synapses, thinned by cryo-fibbing, were observed in the immature and mature state and after pharmacologically induced transmitter release facilitation. Tomography data was analyzed by automated hierarchical segmentation of the network of protein filaments interconnecting synaptic vesicles and by intense statistical evaluation of the overall heterogenous ultrastructure. We found the protein Munc13 to be essential for release facilitation and specific presynaptic phenotypes depending on the activity levels of Munc13 and PKC.
