NEDD8 orchestrates active ubiquitylation assembly of cullin-RING E3 ligase

Abstract: Virtually all cellular processes are regulated by cullin-RING ligases (CRL), the largest family of E3 ligases. CRLs are regulated by a ubiquitin-like protein NEDD8, yet how the modification of NEDD8 catalyzes ubiquitylation remain unknown. Here, the cryo-EM structure of a chemically trapped complex representing the ubiquitylation intermediate by a neddylated CRL1 β-TRCP elucidates how NEDD8 orchestrates the cullin-RING ligase complex in conjunction with the ubiquitin carrying enzyme UBE2D.
