Abstract: For adoptive T-cell transfer of TCR-transgenic T cells, patient-derived T cells are equipped with transgenic TCRs and thereby redirected against tumor cells. Here, we used CRISPR-Cas9 and AAV6 to introduce the transgene specifically into the endogenous TRAC locus, thus circumventing the disadvantages of established semi-randomly integrating vectors. As a result, the surface-expressed TCR is exchanged, and the T cells are functionally redirected against target cells in vitro and in vivo.
