Enhanced Vaccine Adjuvancy via Kinetic Conformational Locking of TLR9 Activation
Year: 2025
Type: article
Abstract: **Abstract:** This research proposes a novel approach to vaccine adjuvancy leveraging kinetic conformational locking of Toll-like Receptor 9 (TLR9) activation. Traditional TLR9 agonists often induce transient activity, limiting immune response durability. Our method utilizes self-assembling peptide polymers engineered to dynamically bind and maintain a specific TLR9 receptor conformation, creating an extended and more robust immune signaling cascade. This enables standardization and amplification of adjuvant effects, leading to improved vaccine efficacy and reduced dosing requirements. The proposed framework, termed "Kinetic Conformational Adjuvancy" (KCA), utilizes dynamic optimization functions for polymer engineering, offering a scalable and customizable solution for enhanced vaccine response across various antigen types.
