IGI.nano-ADx: Programmable Nanosystem for Early, Non-Invasive Detection of Alzheimer's Disease
Year: 2025
Type: article
Abstract: This whitepaper introduces IGI.nano-ADx, a conceptual nanosystem designed to detect prodromal biomarkers of Alzheimer’s disease (AD) through multiplex molecular recognition and non-invasive photoacoustic readout. Unlike current methods, which are invasive (CSF assays) or costly (PET imaging), IGI.nano-ADx targets subtle circulating signals—miRNA panels, hyperphosphorylated Tau species, and neuronal peptide fragments—appearing years before clinical symptoms. The architecture combines: Gold nanorod (GNR@SiO₂) scaffolds with stealth coatings (PEG + zwitterions) for biocompatibility, Three recognition “antennas” (LNA-miRNA probes, pTau-specific nanobodies, neuronal peptide aptamers), Molecular logic (AND gate) requiring co-occurrence of ≥2 biomarkers to reduce false positives, Programmable dimerization of GNRs that shifts plasmonic resonance, generating a strong NIR photoacoustic signal detectable via a skin patch. Expected outcomes: Sensitive, specific detection of Alzheimer’s prodromal stages, Non-invasive, rapid testing (results in 1–6 hours), Potential for population-level screening and longitudinal therapy monitoring, A new paradigm: molecular logic → nanosignal → remote clinical readout.
