Abstract: In recent years, immuno-oncology has made significant strides, fundamentally transforming cancer treatment approaches. Among the array of immunotherapy options, mRNA vaccines have emerged as a groundbreaking modality for combating malignancies an advancement accelerated by their demonstrated success during the COVID-19 pandemic. This review provides a detailed and organized exploration of mRNA-based cancer vaccines, with an emphasis on optimizing their molecular structure, selecting appropriate delivery vehicles, and determining effective routes of administration. We also summarize key findings from both preclinical experiments and clinical investigations to shed light on the present state of mRNA vaccines in oncology. In addition, this review addresses major hurdles hindering the advancement of mRNA cancer vaccines, including both biological limitations and external challenges that impact their clinical application. For mRNA vaccine technology to reach its full potential in oncology, it is essential to deepen our understanding of tumor immunology, expand wellstructured clinical trials, and develop robust, scalable production platforms. Achieving these goals will require cohesive collaboration across disciplines bringing together researchers, healthcare professionals, and industry leaders.
