Tuberculosis Therapeutics: A Multidisciplinary Approach to Developing Novel Drug Regimens
Year: 2025
Type: article
Abstract: Tuberculosis (TB) remains one of the leading causes of death from infectious diseases worldwide, driven in part by the rise of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains. This study synthesizes data from preclinical in vivo murine models, in vitro pharmacological assays, and computational modeling to evaluate two promising anti-tubercular agents, Transitmycin and PA-824. Using established infection models, pharmacokinetic/pharmacodynamic profiling, and predictive treatment-shortening simulations, we assessed bactericidal activity, host-dependent variability, and safety considerations. Both compounds demonstrated significant bacterial burden reduction, favorable pharmacological indices, and potential to shorten treatment duration to under three months while maintaining cure rates above 90%. Transitmycin retained activity in immunocompromised hosts, though dose-dependent toxicity was observed at higher concentrations. These findings highlight the therapeutic potential of these agents in next-generation TB regimens and underscore the importance of integrating computational and experimental approaches to accelerate drug development.
