Smoc1/Mno₂ Np-Loaded Scaffolds For Post-Radiation Bone Regeneration

Year: 2025
Type: article
Abstract: Aim or purpose: ORNJ remains a major clinical challenge. A new antibacterial, antioxidant and osteoinductive bioscaffold has been developed to promote bone regeneration for radiation-damaged bone defects. Materials and methods: The SMOC1(SPARC-related modular calcium-binding protein 1) and Manganese dioxide nanoparticles were loaded into the poly(lactic-co-glycolic acid) /polycaprolactone electrospun scaffold. Cell adhesion, proliferation and differentiation were evaluated using the MC3T3-E1 cell irradiation injury model. And the ability of the scaffolds to promote the repair of radiation-induced bone defects was evaluated using a rat jaw bone defect model of radiation injury. Results: The scaffolds significantly promoted the proliferation of irradiated cells (P < 0.05) and early cell differentiation (P < 0.001). The scaffolds significantly upregulated the expression of BMP2, RUNX2 and OCN genes at both 7 and 14 days by PCR experiments. Cells cocultured with the bioscaffold were found to express more BMP2 and RUNX2 proteins (P < 0.01). These results indicated that the bioscaffolds could effectively promote the differentiation of irradiation-damaged osteoblasts. The bioscaffold also significantly reduced the high ROS levels of the cells (P < 0.01). The bioscaffolds were found to significantly promote new bone regeneration in the bone defect area in the rat jaw. Conclusions: The in vitro experiments showed that the scaffold had good cell compatibility, high osteogenic properties of irradiated cells and ROS scavenging properties, and the in vivo animal experiments showed that the scaffolds promoted bone regeneration. The bioscaffold has potential applications in bone repair caused by radiation injury.
