Effect of Topical Application of Resveratrol Loaded Chitosan Nanoparticles on Chemically Induced Epithelial Dysplasia in Hamster Pouch

Year: 2025
Type: article
Abstract: OBJECTIVES: This research aimed to investigate the topical effect of Resveratrol (RV) loaded on chitosan nanoparticles (CS-NP) on chemically induced epithelial dysplasia in the hamster buccal pouch. MATERIAL AND METHODS: 35 Syrian hamsters were used in this study; they were divided into 4 groups. Group A (n = 5) was the negative control group. 7,12-dimethylbenzo anthracene (DMBA) was applied topically for 6 weeks to induce oral epithelial dysplasia in the remaining 30 animals, then they were divided into group B (n = 10) as the positive control group, group C (n = 10) animals received CS-NP (0.85 mg/100 L) topically, and group D (n = 10) animals received RV-CS-NP (0.85 mg/100 L) topically for another 6 weeks. Serial sections of the buccal pouches from all animals were examined histologically, and immunohistochemical (IHC) detection of the Bcl-2 gene expression was assessed. RESULTS: The results showed that treatment with RV loaded on CS-NPs had significantly reduced the incidence of epithelial dysplastic lesions induced by the carcinogen. Data analysis showed statistically significant differences (p < 0.001) in the expression levels of Bcl-2 through IHC between the treatment and positive control groups. CONCLUSION: Topical resveratrol-loaded chitosan nanoparticles are promising chemopreventive therapy.
