Abstract: In this issue of Blood, Meaker et al 1 report that the transcription factor Runx2 is an important negative regulator of hematopoietic stem cell (HSC) expansion and T-cell lineage commitment ex vivo and in vivo through the use of a genome-wide CRISPR knockout (KO) screen in HSCs, improving our understanding of intrinsic factors that control HSC proliferation and cell fate.
