109P CTO1681 does not interfere with CAR T cell anti-tumor efficacy in vivo

Year: 2025
Type: article
Abstract: CD19-directed chimeric antigen receptor (CAR) T-cells have transformed outcomes for relapsed or refractory B-cell malignancies, achieving complete-response rates of 50–80%. Despite this transformative care, broader adoption and outpatient use of CAR T-cell treatments are constrained by the development of potentially life-threatening cytokine-release syndrome (CRS) and immune-cell associated neurotoxicity syndrome (ICANS) which require inpatient management. Current management of CRS/ICANS involves the use of tocilizumab or high-dose corticosteroids.
