Apigenin and Eupatilin: A Pharmacognostic and Pharmacological Perspective

Year: 2026
Type: article
Abstract: :Apigenin (4′,5,7-trihydroxyflavone) and Eupatilin (5,7-dihydroxy-3′,4′,6-trimethoxyflavone) arenaturally occurring flavones with broad pharmacological significance. They are primarilyobtained from Matricaria chamomilla, Petroselinum crispum, Citrus aurantium, and Artemisia asiatica. Both compounds possess diverse biological activities, including antioxidant, anti-inflammatory, anticancer, neuroprotective, hepatoprotective, and gastroprotective effects. This chapter presents a detailed pharmacognostic, phytochemical, and pharmacological profile ofApigenin and Eupatilin, focusing on their structural characteristics, molecular mechanisms, andtherapeutic relevance. The pharmacognostic section discusses botanical sources, morphologicalfeatures, and optimized extraction and isolation procedures involving ethanol, methanol, andchromatographic purification. Phytochemical characterization includes spectroscopic data suchas UV, IR, NMR, and MS confirming hydroxyl and methoxy functional groups. Quantitativeestimation through HPLC and HPTLC ensures standardization and quality control of plantextracts containing these flavones. Apigenin acts by modulating NF-κB, STAT3, and MAPKpathways, leading to the suppression of pro-inflammatory mediators, inhibition of tumor cellproliferation, and enhancement of neuronal protection. Eupatilin mediates its effects primarilythrough inhibition of COX-2, TGF-β, and activation of the Nrf2/HO-1 antioxidant system, contributing to gastroprotection and cytoprotection against oxidative damage. Both compoundsexhibit significant free radical scavenging and anti-apoptotic activities in preclinical models.Pharmacokinetic studies reveal limited solubility and low oral bioavailability due to extensiveglucuronidation and sulfation. Novel drug delivery approaches such as nanoparticles, liposomes, and cyclodextrin inclusion complexes have improved their pharmacokinetic profiles.Toxicological data confirm high safety margins without organ-specific toxicity. Recent researchhighlights advances in nanoformulations, synergistic applications, and targeted delivery systems.Despite promising preclinical results, clinical validation remains limited, emphasizing the needfor further mechanistic and pharmacokinetic optimization. The comprehensive pharmacognosticand pharmacological evaluation presented in this chapter supports Apigenin and Eupatilin aspotential lead molecules for future phytopharmaceutical and therapeutic development.
