Abstract: Background/Aim: Autogenous grafts have many drawbacks as donor site morbidity and postsurgical complications. This study aimed to find an efficient substitute for autogenous grafts by evaluating the biodegradation and remodeling of a novel naturally derived scaffold with added (rhPDGF-BB) for volume augmentation, regeneration and reconstruction of soft tissue defects.Material and methods: Two types (groups) of bovine acellular dermal collagen matrices (ADCM) were isolated and grafted to cover the skin defects on the dorsum of 14 male Sprague–Dawley rats. Before grafting, half of them were loaded with rhPDGF-BB and the other were used as control. Histological, immunohistochemical and histomorphometric analysis were performed 2 and 4 weeks after grafting. ResultsBoth groups showed proper soft-tissue integration. At 2 weeks, enriched scaffolds demonstrated a marked increase in fibrovascular tissue compared with controls, a difference that became less pronounced by 4 weeks. Vimentin staining confirmed fibroblast activity in both groups, with higher expression in enriched scaffolds. Overall, longer healing periods enhanced regeneration, with ADCM2 outperforming ADCM1. ADCM2 showed earlier and more active soft-tissue regeneration at 2 and 4 weeks, maintained superior outcomes through greater fibroblast presence, richer fibrovascular networks, and more organized collagen deposition. These results indicate that ADCM2 offers a more favorable environment for periodontal soft-tissue regeneration and augmentation. Conclusions: The addition of (rhPDGF-BB) to a native bovine (ADCM) facilitated vascularization, connective tissue formation and tissue integration of novel scaffolds and helped to use it as a promising alternative to autogenous grafts for regeneration and augmentation of oral soft tissue defects.
