A Deterministic Composition→Optical→Electromagnetic Closure: Coene–Cocito Double-Label %GC Lifted into the Maxwell–Scretching Chain Using Revised NMP Extinction Coefficients

Abstract: This work presents a deterministic, first-principles framework that bridges enzymatic DNA base-composition measurement and electromagnetic optical theory within a single, closed mathematical chain. Starting from the Coene–Cocito double-label nick-translation method, which determines DNA base composition (%G+C) from an isotope incorporation ratio, the study rigorously verifies the underlying stoichiometric estimator and its required correction conditions. Using the revised nucleoside-5′-monophosphate ultraviolet extinction coefficients reported by Cavaluzzi & Borer (2004), the %G+C result is lifted into the Maxwell–Scretching Chain, yielding an explicit and invertible mapping from composition to molar absorptivity at 260 nm and onward to macroscopic electromagnetic state variables. The resulting closure links the isotope ratio directly to absorbance, absorption coefficient, complex refractive index, imaginary permittivity, and effective conductivity, with all steps expressed as numbered equations and without empirical fitting. This deterministic composition→optical→electromagnetic closure provides a new calibration bridge between biochemical composition assays and UV–visible spectrophotometry, enabling reference-grade DNA quantitation and electromagnetic state inference even in regimes where classical buoyant-density or melting-temperature correlations fail. The framework is immediately applicable to molecular biology, biophysics, spectroscopy, and instrument validation, and serves as a foundational component of the broader Maxwell–Scretching and Scretching Quantum Chain theories.
