Influence of three chemical disinfection protocols on the mechanical, physical, and cytotoxic properties of additively manufactured complete denture bases with and without soft reline resin: An in vitro study

Abstract: STATEMENT OF PROBLEM: Additive manufacturing using stereolithography (SLA) has emerged as a promising method of manufacturing complete denture bases. However, patients wearing these prostheses often require tissue conditioning and regular disinfection. While chemical disinfectants have been widely used in clinical practice, their effects on the mechanical properties and cytotoxicity of relined SLA-manufactured dentures are not well established, and a standardized disinfection protocol is unavailable. PURPOSE: The purpose of this in vitro study was to evaluate the effects of 3 chemical disinfection protocols on the mechanical, physical, and cytotoxic properties of complete denture base resins manufactured by SLA, with or without resilient resin. MATERIAL AND METHODS: A total of 168 specimens were prepared and divided into 6 groups (n=14 per condition). SLA-manufactured denture base specimens (Denture Base; Formlabs) were either left nonrelined (SLA-NR) or relined with a soft tissue-conditioning acrylic resin (GC Tissue Conditioner; GC) (SLA-R). The disinfection protocols were 2% glutaraldehyde (2 minutes), 0.3% peracetic acid (1 minute), and alkaline peroxide-based effervescent tablets (3 minutes), applied in 6 cycles. Control groups had no disinfection. Cytotoxicity and mechanical and physical properties were assessed with 3-point flexural strength, fracture toughness, surface roughness, Vickers hardness, and Shore A hardness. Statistical analysis included normality testing (Kolmogorov-Smirnov), ANOVA with Tukey post hoc tests for parametric data, and Kruskal-Wallis with Dunn post hoc tests for nonparametric data (α=.05). RESULTS: ), and surface roughness (Ra: 3.95 ±0.36 to 4.44 ±0.35 µm) were statistically similar (P>.05). All relined specimens showed 0% cell viability, while nonrelined specimens maintained 98% to 100% viability (P<.001). CONCLUSIONS: Among the tested protocols, glutaraldehyde best preserved the mechanical integrity of SLA-relined dentures.
