494eP Pharmacogenomic biomarkers and CNS progression in EGFR-mutant NSCLC patients treated with first-line osimertinib: A retrospective cohort study
Year: 2026
Type: article
Study design: Observational Study
Abstract: Osimertinib is a third-generation EGFR-TKI and a substrate of the ABCB1 and ABCG2 efflux transporters, which restrict its CNS penetration. Specific SNPs (e.g., ABCB1 3435C>T, ABCG2 34G>A) may impair transporter function and have been associated with increased risk of CNS progression, possibly due to reduced intracerebral drug exposure.
