Abstract: Lipid species in tumour spheroids resemble those found in orthotopic breast xenograft tumours. (a) BT474 were cultured in normoxia (20 % O2) or hypoxia (0.1 % O2) for 48hrs before lipids were extracted and used to determine the relative amount of different diacylglycerol (DG) species. Archival tissue from six individual tumours was also analysed. (b) Relative distribution of phosphatidylcholine (PC) species in BT474 cells grown under different conditions. Data represent the mean ±SEM of three biologically independent replicates. Statistical comparisons were performed using Student t-test (*p≤0.05 compared to 2D hypoxia, #p≤0.05 compared to 2D normoxia). (c) Relative distribution of different DG species in T47D cells grown in 2D or as tumour spheroids. (d) Relative distribution of different monoacylglycerol (MG) species in T47D cells grown as in c. Data represent the mean ±SEM of three biologically independent replicates. Statistical comparisons were performed using Student t-test (*p≤0.05). (PDF 533 kb)
