Methodological limitations in interpreting the efficacy of trastuzumab deruxtecan by CLDN18.2 status in single-arm studies
Year: 2026
Type: article
Abstract: We read with great interest the article by Okemoto et al. investigating the association between claudin 18.2 (CLDN18.2) expression and clinical outcomes of trastuzumab deruxtecan (T-DXd) in patients with human epidermal growth factor receptor 2 (HER2)-positive gastric or gastroesophageal junction cancer.1 While the study addresses a clinically significant question, we would like to highlight several methodological concerns that may affect the interpretation of the findings.
