73P Enhancing CAR T cell therapy through the epigenetic reprogramming of memory gene transcriptional circuits

Year: 2026
Type: article
Abstract: Chimeric Antigen Receptors (CARs) are synthetic constructs that combine the effector functions of T cells with the antigen-binding specificity of antibodies. Extensive research has highlighted the impact of epigenetic mechanisms in regulating T cell differentiation, identifying the histone methyltransferase EZH2 as a key determinant. As the catalytic subunit of the Polycomb Repressive Complex 2, EZH2 mediates the trimethylation of histone H3 at lysine 27, a mark associated with gene repression. Transcriptomic analyses of CAR T cells have shown that memory-associated gene expression correlates with durable responses and persistence, whereas exhaustion-related programs are linked to poor outcomes.
