122eP Preclinical investigation of the potential synergistic action of small molecule inhibitors in combination with chemotherapy in colon and liver cancer cells
Year: 2026
Type: article
Abstract: Colon and liver cancers are among the most prevalent malignancies worldwide in both sexes. Cisplatin is a widely used chemotherapeutic agent; however, its clinical effectiveness is limited by the development of resistance and toxicity. Cisplatin acts primarily through DNA crosslinking, resulting in DNA damage, cell-cycle arrest, and apoptosis. Enhancing its therapeutic impact through combination strategies, allows for lower effective dosage. Targeting the PI3K/AKT/mTOR pathway, a key regulator of cell survival often upregulated in cancer, using small-molecule inhibitors (SMIs) has shown promise.
