Abstract: Yeast surface display sorting of trastuzumab variant library for identifying HALA antibodies and sequence of lead clone (S12). (A) Two gates of antigen-binding antibodies (R6 and R7) were collected to isolate antibody variants with different binding properties. (B) Binding characterization of selected mutants on the yeast surface using monovalent (HER2-mFc) and bivalent (HER2-Fc) HER2. (C) The S12, S18, and M11 clones were sequenced and contain the same VL domain as trastuzumab. The VH and VL domains are shown with CDRs underlined.
