Four-Feature AND-Gate Cancer Selectivity: A Framework for Early-Stage Cancer Interception via Mesoporous Silica Nanoparticle Delivery
Year: 2026
Type: preprint
Abstract: This hypothesis paper proposes a four-feature AND-gate cancer selectivity framework requiring four independent biological features—phosphatidylserine exposure, Thomsen-Friedenreich antigen expression, elevated reactive oxygen species, and active telomerase—to be satisfied simultaneously for cytotoxic activation. Implementation via mesoporous silica nanoparticle platform with bispecific PSBP-6/JAA-F11 surface targeting, CD47 mimetic anti-phagocytic protection, ROS-cleavable linker, and telomerase-responsive DNA hairpin payload uses substantially established chemistry. The framework extends recent work by Yilmaz et al. (2024) on phosphatidyl-conjugated telomerase-targeted cancer killing and Shi et al. (2018) on telomerase-responsive DNA hairpin release, integrating these with additional selectivity features through orthogonal biological dimensions. Coverage estimates of 63-80% across cancer types represent a deliberate selectivity-coverage trade-off favoring precision over breadth. The contribution is configurational rather than mechanistic, with explicit falsification conditions and acknowledged limitations including long-term safety unknowns and required chemistry development.
