Generation of WTC11 CLYBL-CAG_GCaMP8f cell line for fast and sensitive calcium tracking

Abstract: Genetically encoded calcium indicators (GECIs), such as GFP-based GCaMP sensors, are powerful tools for monitoring electrical activity in excitable cells. Here, we engineered a WTC11 line by inserting the fast GCaMP8f sensor into the CLYBL safe-harbor locus under the control of the CAG promoter, together with an antibiotic selection cassette removable by Cre recombinase. Cardiomyocytes and neurons differentiated from this line express the reporter robustly, with GFP fluorescence dynamics closely tracking cellular calcium. This cell line provides a versatile and reliable platform for investigators requiring rapid calcium monitoring with preserved sensitivity and brightness.
