Abstract: This collection comprises 48 datasets generated through 24 paired PhenoCycler-Fusion proteomics and 24 high-resolution H&E histology datasets derived from profiling of human pancreatic tissue, aimed at characterizing the distribution and biological heterogeneity of senescent cells and their associated secretory phenotype across the adult lifespan. Tissue sections were derived from two or more anatomical regions — head superior, head inferior, middle, and tail — from whole pancreases collected from brain-dead donors. PhenoCycler-Fusion (Akoya Biosciences) profiling using a custom antibody panel provides spatially resolved protein expression data, enabling the identification of cell types within key anatomical structures including ducts, vasculature, and islets of Langerhans. The same slides were subsequently stained with hematoxylin and eosin (H&E) and digitized at 0.22 µm per pixel, generating whole slide images used for anatomical annotation, automated machine learning-based tissue and nuclear segmentation, and morphological quantification. Importantly, these whole slide images also serve a guiding role, informing the selection of regions of interest for complementary profiling of adjacent tissue sections with additional assays. Together, these two modalities provide spatially resolved proteomic and morphological characterization of the healthy human pancreas in the context of aging and cellular senescence.
