Abstract: Supplemental Fig. S10. The combination of Trametinib and anti-PD-1 antibody attenuates the immunosuppressive TiME of the KPC2138-FIO tumors. A-B. Mice with established orthotopic 2138-FIO (A) and subcutaneous (B) tumors were randomized and treated with Trametinib (1 mg/kg, daily) or vehicle control in combination with an anti-PD-1 antibody (250 μg, three times/week) or a control isotype-matched IgG antibody (250 μg, three times/week). After 2 weeks, tumors were collected for analysis by flow cytometry for the frequency of all tumor-associated macrophages (TAM) (CD45+CD11b+F480+), M2 (CD45+CD11b+F480+Arg1+), and M1 TAMs (CD45+CD11b+F480+MHCII+). C. Subcutaneous KPC2138-FIO tumors at endpoint were also evaluated for the expression of select immunosuppressive markers (CD206, PD-L1, Arginase1 (Arg1)) on granulocytic myeloid-derived suppressive cells (G-MDSCs; CD45+CD11b+Ly6G+Ly6C-), monocytic myeloid-derived suppressive cells (M-MDSCs; CD45+CD11b+Ly6G-Ly6C+), and TAMs. One-way ANOVA, followed by Tukey multiple comparison test. *, p<0.05 **, p<0.01 ***, p<0.001 ns, non-significant.
