Abstract: Figure S16. Stability assessment of [225Ac]Macropa-PEG4-YS5-MMAE over 7-day storage in PBS. (A) Drug-to-antibody ratio (DAR) was measured on Day 0, Day 4, and Day 7 using UV spectrophotometry. DAR remained stable between 2.2 and 2.8, confirming MMAE retention over time. (B) Magnetic bead–based binding assay performed on Day 7 demonstrated preserved immunoreactivity of [225Ac]Macropa-PEG4-YS5-MMAE to CD46 antigen. Binding was significantly reduced in the presence of excess unlabeled YS5 (blocking group) or in the absence of CD46. (C) CellTiter-Glo viability assay using 22Rv1 cells treated with [225Ac]Macropa-PEG4-YS5-MMAE on Day 0 or after 7 days of storage in PBS showed no significant difference in cytotoxic activity, confirming functional stability.
