Multiscale Study of the Astrocyte-Synapse Interface: From Light Microscopy to Cryo-ET

Year: 2026
Type: dataset
Abstract: Astrocytes regulate neuronal circuits through dynamic interactions with synapses. Recent work (accepted in Cell) reveals that fine astrocytic processes form leaflet domains interconnected by gap junctions, contacting multiple synapses and enriched in ER and ribosomes. These domains host localized calcium events independent of mitochondria and IP₃R2, but sensitive to IP₃R1 deletion, suggesting they act as hubs coordinating synaptic activity beyond the classical tripartite synapse. To investigate them, we propose a correlative cryogenic imaging strategy combining: (i) immunofluorescence to locate astrocytic and synaptic clusters, (ii) cryo-X-ray nano-holotomography and fluorescence mapping at ID16A, and (iii) serial lift-out with cryo-electron tomography at CM02. This pipeline will map leaflet domains from light to nanometer resolution, correlate molecular markers with ultrastructure, and uncover their role in neuroglial communication across ESRF and IBS.
