Abstract: A) PCR amplification of the LSM1-FGFR1 fusion and breakpoint sequence identified in an osteosarcoma patient (zcc386). B) Cell number and viability of LSM1-FGFR1 and OP2-FGFR1-expressing Ba/F3 cells following IL-3 withdrawal over ten-day time course. Total number of viable cells was determined by trypan blue exclusion. Data presented as mean ± standard error of the mean (SEM) (n=3). C) Viability analysis of LSM1-FGFR1- and FGFR1OP2-FGFR1 (OP2-FGFR1)-expressing Ba/F3 cells following 96-hour (left) and 168-hour (right) IL-3 withdrawal. Viability was determined by propidium iodide exclusion, measured by flow cytometry. FGFR1 variant cell lines were compared to MSCV controls using a one-tailed unpaired t-test (*: P < 0.05, ****: P < 0.0001). Data presented as mean ± SEM (n=3). ¬¬D) Viability analysis of LSM1-FGFR1- and OP2-FGFR1-expressing Ba/F3 cells treated with a dose titration of regorafenib, AZD4547 or imatinib. Data presented as mean ± SEM (n=6).
