Engineered Bacteroides thetaiotaomicron for Targeted Elimination of Fusobacterium nucleatum and Prevention of Colorectal Cancer

Year: 2026
Type: article
Abstract: The S100 Therapeutic Pill is a novel engineered Bt-based therapeutic for targeting F. nucleatum and preventing colorectal cancer. The design includes 4 genomic edits: pyrE deletion (uracil auxotrophy), tdk replacement with SQR/FRD operon (H₂S detoxification), dnaE TAG at codon #2 (ncAA auxotrophy), and ompA thinning (hypervesiculation). The pill formulation includes sodium fumarate, uracil, and a non-canonical amino acid, with a pH-responsive coating for colonic release. Absolute biocontainment with 10⁻³² escape probability.
