A structure-guided hypothesis for sequential S8/PtpA cleavage of the Ara h 2 DPYSPS loop
Year: 2026
Type: preprint
Abstract: An outcome-blind computational hypothesis and laboratory-ready blinded validation protocol. The work proposes that an S8/S53-family endoprotease may expose a QDP-starting Ara h 2 neo-N terminus at S68|Q69 or S75|Q76, followed by S9B/PtpA-like removal of QDP. It provides exact linked product signatures, four coequal same-assembly candidate pairs, public evidence tables, and separate operations and analyst-blinded assay packets. No candidate enzyme activity, sequential cleavage, reduced IgE binding, therapeutic benefit, clinical safety, or human dosing has been demonstrated.
