Abstract: Objective This study aimed to investigate whether the auto-registration feature of improved holographic imaging technology improves the accuracy and efficiency of prostate biopsy. Methods Six healthy adult male Labrador retrievers were randomized into observation and control groups. Calcium carbonate-agar composite gel was implanted into the canine prostate to create mimic lesions. In the observation group, repeat implantation at the original lesion mimic site was performed under improved holographic imaging guidance to simulate targeted prostate biopsy, while the control group underwent the same procedure using conventional holographic guidance. All animals underwent Whole-abdomen CT scans followed by prostatectomy for whole mount sections examination. The positional differences of the lesion mimics implanted twice in the prostate of the two groups of dogs in three-dimensional space were compared respectively on CT images and large tissue sections. Procedure times for repeat implantation were recorded and compared between two groups. Results On CT images and whole mount sections, the distances of the lesion mimics implanted twice in the control group along the X-axis, Y-axis, and Z-axis were 4.42 ± 1.25 mm & 4.05 ± 1.18 mm, 2.61 ± 1.15 mm & 2.44 ± 1.32 mm, and 3.88 ± 1.42 mm & 3.58 ± 1.51 mm, respectively. The observation groups were 1.94 ± 1.43 mm & 1.71 ± 1.19 mm, 1.62 ± 1.27 mm & 1.28 ± 1.33 mm, and 1.82 ± 1.35 mm & 1.57 ± 1.22 mm, respectively. Whether on CT images or whole mount sections, the distances of the lesion mimics implanted twice in the control group along the X-axis, Y-axis, and Z-axis were significantly longer than those in the observation group ( P < 0.05). The time required from the fixation of the experimental dog's position to the completion of implantation in the control group was 18.76 ± 2.59 min, while that in the observation group was 11.32 ± 1.75 min, which was significantly shorter than that in the control group ( P = 0.015). Conclusion The automatic registration function of the improved holographic imaging technology can, to a certain extent, shorten the operation time while improving the accuracy of puncture.
