Year: 2026
Type: article
Abstract: Engineering a High-Concentration Spermidine Formulation: From Molecular Biology to Pharmaceutical Formulation Design and Development This publication presents a comprehensive review of the pharmaceutical engineering principles involved in the development of a high-concentration aqueous spermidine formulation. Rather than focusing solely on the biological activity of spermidine, the review examines how formulation architecture, physicochemical optimization and pharmaceutical engineering collectively determine formulation quality and the preservation of molecular integrity. The manuscript discusses the scientific relationship between molecular biology and pharmaceutical formulation science, with particular emphasis on concentration-dependent formulation challenges, including pH optimization, osmolality, molecular stability, excipient selection and long-term physicochemical robustness. A representative formulation containing spermidine and trehalose is used as a case study to illustrate the engineering principles underlying advanced longevity formulations. The review introduces the concepts of Bioengineered Formulation Architecture (BFA) and Precision Longevity Formulation Engineering (PLFE), describing an interdisciplinary framework that integrates molecular biology, pharmaceutical engineering, physical chemistry and formulation science to support the rational design of next-generation longevity formulations. Topics covered include: Spermidine biology, autophagy and mitophagy Pharmaceutical formulation engineering High-concentration aqueous formulation design Physicochemical optimization pH and osmolality engineering Stability engineering and Quality by Design (QbD) Trehalose as a multifunctional formulation component Bioengineered Formulation Architecture (BFA) Precision Longevity Formulation Engineering (PLFE) This publication is intended for educational and scientific purposes and aims to contribute to the growing interface between pharmaceutical engineering, formulation science and longevity biomedical research. It does not constitute medical advice or therapeutic recommendations.
