Abstract: (A) Dose-response curves represent ziftomenib sensitivities of HOXA-low vs HOXA-high T-ALL cell lines. (B) Heatmap showing RNA expression of HOXB, HOXC, HOXD genes in T-ALL patient samples. (C) Splenectomy specimens of NSG mice engrafted with KARPAS-45 cells and treated with vehicle vs ziftomenib at two different dose levels for 4 weeks. (D,E) Tumor burden in bone marrow and spleen of treated mice. (F) Histopathologic analysis of organs harvested from NSG mice treated with vehicle or ziftomenib at two different dose levels showed increased leukemic cell burden (arrows) in livers of vehicle-treated mice but no microscopic evidence of toxicity to liver, heart, kidney or guts.
