Understanding the tolerance of chromosomal instability under epithelial to mesenchymal transition in cancer

Type: dissertation
Abstract: Cancer remains a major global health challenge, primarily because the underlying mechanisms are still poorly understood. In recent years, research has shifted its focus from studying molecular pathways to examining the hallmarks of cancer, key traits that enable cells become malignant, being genomic instability one of the most important in this progression. Understanding how this genomic instability appears is essential to avoid cancer development and drug resistance. Here we show karyotyping analysis, immunofluorescence and immunoblotting assays, in addition to DNA content profiles to elucidate the role of epithelial-mesenchymal transition (EMT) using TGF-β treatment in the appearance of genomic instability. Additionally, we tried to develop new tumorigenic cell lines from immortalized epithelial cells through soft-agar growth experiments. Our results show that epithelial cells increase their tolerance to DNA damage under EMT condition and that p21 and p53 are less activated under these conditions, explaining such tolerance. However, further experiments should be conducted to validate these results.
