Regulation of innate immune cell differentiation by mitochondrial nicotinamide adenine dinucleotide (NAD+) levels

Abstract: Macrophages can be divided roughly into two types in vitro: M1 or proEinflammatory, that display a glycolytic metabolism, and M2 or reparative, that largely rely on oxidative phosphorylation. Nicotinamide adenine dinucleotide (NAD+) is an essential cofactor in redox metabolism and metabolic signaling. To characterize the effect of mitochondrial NAD+ levels in the polarization of macrophages, we studied both the silencing and overexpression of MCART1 (SLC25A51), the mitochondrial NAD+ transporter (1) , in THPE1 cells and mouse bone marrow derived macrophages (BMDM).
