Abstract: PSMAi-C’ dots modulate tumor and immune markers and upregulate interferon regulatory factors (IRFs) in the Hi-Myc model, with or without immune checkpoint blockade. (a, b) Gene expression profiles of tumor tissues harvested from animals 4 days after the final dose of a multi-dose (three doses every three days) regimen of i.v.-injected saline vehicle, ICB, PSMAi-C’ dots, or Dual Tx (n = 3 mice/cohort). Significant fold changes are indicated by values above 2, demarcated by dashed lines. (c-f) Representative tumor tissue specimens from all treatment arms were stained by (c) IHC and quantified for (d) cMyc, (e) Ki-67, and (f) PD-L1 (scale bar: 250 um). All numerical data are presented as mean ± s.e.m. 1-way ANOVA with Kruskal-Wallis test was performed in (a,b) and (d-f). *p < 0.05, **p < 0.01.
