Abstract: Particle treatment in combination with CSF-1R blockade reduces suppressive myeloid populations and improves treatment response in prostate cancer models. (a) Tumor growth was monitored over a 28-day period post-implantation in Myc-CaP tumor-bearing animals. A multi-dose regimen (three doses every three days) of one of the following treatments (n = 4 treatment arms, 3 animals/arm) was initiated 7 days post-implantation: (i) saline vehicle, (ii) PSMAi-C’ dots (12 nmol/dose), (iii) PSMAi-C’ dots + i.p.-injected anti-CD8 (αCD8), and (iv) PSMAi-C’ dots + i.p.-injected anti-CSF-1R depleting antibodies (αCSF-1R). (b) Antibody-mediated CD8+ T cell depletion confirmed by flow cytometry with histograms (upper panel) and bar plots (lower panel) depicting the percentages of cells in blood after the first and final particle doses using the same treatment strategy as in (a). (c – f) Bar plots depict the percentage of distinct myeloid cell populations derived from harvested tumors 4 days after the final administered dose by flow cytometry using the same treatment strategy as in (a). (g) Schematic illustrating isolated BMDMs from untreated Hi-Myc tumor-bearing mice that were treated ex vivo with vehicle, PSMAi-C’ dots, αCSF-1R, and a combination of PSMAi-C’ dots + αCSF-1R. (h – j) Percentage of myeloid cell populations analyzed across treatment groups by flow cytometry 48 h after particle exposure. All data are presented as mean ± s.e.m (n = 3 mice/treatment arm). 1-way ANOVA with Tukey's multiple comparisons test was performed in (a, c – f) and (h – j). Student’s t-test was performed in (b). *p < 0.05, **p < 0.01, ***p < 0.005, ****p < 0.001, ns, not significant.
