Abstract: Particle-based therapy reprograms myeloid phenotypes, promotes phagocytosis, and boosts T cell–mediated immune activation in Hi-Myc splenocytes. (a) Schematic depicting the timeline of Hi-Myc tumor development in positively genotyped mice, along with the experimental procedure for isolating/culturing bone marrow-derived macrophages (BMDMs) and splenocytes (T cells) for ex vivo analyses following treatment of mice with saline, PSMAi-C’ dots, ICB, or Dual Tx. (b) Percentage (%) of tumor cells phagocytosed by co-cultured BMDMs using flow cytometry 24 h after exposing the former to treatments specified in (a). Percentage of cells co-expressing F4/80 and CFSE were gated to identify phagocytic macrophages. (c) Fold changes of representative genes in BMDMs exposed to treatments in (a) for 48 h and assayed via RT-qPCR. (d – g) ELISpot assay of IFN-γ, TNF-α, and IFN-γTNF-α secretion from isolated splenocytes (a). Data in (b, c, e – g) are presented as mean ± s.e.m. and a 1-way ANOVA with Brown-Forsythe test was performed. *p < 0.05, **p < 0.01, ****p < 0.001.
