Abstract: Particle-mediated ferroptotic induction remodels the Myc-CaP TME to an anti-tumor proinflammatory phenotype (Day 7 post treatment). (a) Myc-CaP flank tumor-bearing mice were assigned to one of three treatment arms (n = 3/arm): saline vehicle, PSMAi-C’ dots, and PSMAi-C’ dots plus liproxstatin-1 (LP-1). Tumor growth was monitored for 7 days after the final treatment. (b) Representative H&E and c-myc staining of ex-vivo tumor sections from representative mice in (a); scale bar: 500 um. (c) Video highlighting differences in live untreated cells and dead (Sytox Green+) Myc-CaP cells using treatments in (a). IHC analyses of tumors harvested from (a) illustrating the percentage of (d) CD3+, (e) CD4+, (f) CD8+ T cells, (g) Iba1+ macrophages, and (h) c-myc + cells (n = 3–6 /arm). All numerical data are presented as mean ± s.e.m. 1-way ANOVA with Tukey's multiple comparisons test was performed for (d – h). *p < 0.05, **p < 0.01, ***p < 0.005, ****p < 0.001.
