Strain-level Diversity in Streptococcus mutans Virulence Traits - Quantification of Isolate Biofilms

Year: 2026
Type: dataset
Abstract: This dataset contains the raw and processed experimental data supporting a study of strain-level phenotypic diversity among genetically diverse clinical isolates of Streptococcus mutans, an important bacterial contributor to dental caries. The study examines how individual S. mutans strains differ in biofilm architecture, extracellular matrix composition, responses to human saliva, growth and biofilm formation under acidic conditions, acid-associated microenvironments within mature biofilms, and susceptibility to antimicrobial agents commonly used in oral healthcare. The deposited data include microscopy images and quantitative measurements of biofilm microcolony size and organization, cellular biomass, glucan and extracellular DNA accumulation, growth measurements under different starting pH conditions, crystal violet biofilm biomass assays, pHrodo-based measurements of relative acid-associated fluorescence within biofilm microcolonies, antimicrobial biofilm inhibition assays using chlorhexidine and cetylpyridinium chloride, and membrane-integrity measurements following short-term chlorhexidine exposure.Together, these datasets document substantial functional diversity across S. mutans strain backgrounds and demonstrate that biofilm architecture, acid-associated physiology, acid tolerance, and antimicrobial susceptibility are only partially coupled. The collection is provided to facilitate reproducibility, secondary analysis, and comparative studies of strain-level variation in S. mutans and cariogenic biofilm biology.
