Abstract: This collection comprises 92 datasets generated through sequential multimodal profiling of human pancreatic tissue, combining 32 Xenium In Situ transcriptomics datasets, 28 PhenoCycler-Fusion proteomics, and 32 datasets of high-resolution H&E histology to characterize the distribution and biological heterogeneity of senescent cells across the adult lifespan. Tissue sections were derived from two or more anatomical regions — head superior, head inferior, middle, and tail — from whole pancreases collected from brain-dead donors, with each modality applied to the same slides in succession. Xenium In Situ (10x Genomics) profiling using a custom 300-gene v1 panel provides subcellular-resolution spatial transcriptomics, enabling the identification of spatially resolved cell types within key anatomical structures such as ducts, vasculature, and islets of Langerhans. The same slides were subsequently profiled with PhenoCycler-Fusion (Akoya Biosciences) using custom antibody panels to capture spatially resolved protein expression across the same tissue regions. Finally, hematoxylin and eosin (H&E) staining was performed on the processed slides and digitized at 0.137 µm per pixel, generating whole slide images used for anatomical annotation and automated machine learning-based tissue and nuclear segmentation for morphological quantification. Together, these three modalities form a deeply integrated spatial atlas of the healthy human pancreas, purpose-built to map senescence and its associated secretory phenotype at cellular and subcellular resolution.
