Abstract: CD8+ and CD4+ T cells from lungs were sorted to at least 99% purity using a BD FACSAria III and single-cell suspensions were loaded onto a Chromium Single-Cell Controller instrument (10x Genomics) to generate single-cell gel beads in emulsion (GEMs). Single-cell RNA libraries were prepared using the Chromium Single-Cell 3’ Library v3 and Gel Bead Kit. Libraries were constructed using the Chromium single-cell library construction kit.
