Abstract: Mature bovine adipocytes were cultured in basal medium (CON) for 7 d. On d7, adipocytes were challenged with hydrogen peroxide (H2O2; 300 μM) for 0.5, 2, and 4 hr. BAS (no H₂O₂; unchallenged control) served as the baseline reference for challenge comparisons. (A) Viability is expressed as Calcein AM fluorescence (RFU). Saponin (0.1%, 10 min) was used to generate a dead cell control. (B) Integrated green fluorescence intensity was used to quantify caspase-3 activity. Doxorubicin (Dox; 10 µM, 3 h) served as a positive control for apoptosis induction. Data are represented as mean ± SEM. One-way ANOVA followed by Tukey’s post hoc test was used to compare treatment groups. Bars with different letters (a, b, c) are statistically significant (P ≤ 0.05).
