Abstract: This study investigates the molecular organization of glutamate receptors within hippocampal excitatory synapses. Using approximately tenfold tissue expansion combined with STED microscopy, we mapped endogenous AMPA receptors, NMDA receptors and PSD-95 at near-receptor-complex scale. The data show that mesoscale receptor nanodomains can arise without discrete multi-receptor clustering and support a model in which receptor-scale PSD-95-associated scaffold units provide a scalable architecture for regulating receptor abundance and composition during postnatal maturation and learning.
