SPAN: a geometric model of effector tethering in R-loop genome editors

Year: 2026
Type: software
Abstract: SPAN computes where a deaminase tethered to a CRISPR effector can act on the displaced strand of an R-loop, from linker statistics, steric exclusion and R-loop geometry alone. Three parameters are fitted to two previously reported editing profiles and held fixed; all further predictions are made without adjustment and are scored with each substrate position's deposited coordinate withheld. The deposit contains the model, the analysis scripts, the results produced by those scripts, the container definition, and a corpus of 50 architectures from seven studies, with a source register that provides the status of each study examined. Structural coordinates are not redistributed; a script retrieves them by accession from the Protein Data Bank.
